A Study on Transcriptional Response of a Glutathione S-transferase Gene of Physarum polycephalum to Nano-sized TiO2

Abstract:The widespread use of nano-sized TiO2 (nTiO2) and releasing into the environment are very likely to cause pollution so there is an urgent need to develop a biomarker for nTiO2 exposure. In this paper, the stress of nTiO2 with various diameters (25, 43, 67 nm) on an environmentally sensitive eukaryotic slime molds Physarum polycephalum was studied. It was found that the total enzyme activity of GST was increased by 8~44 fold with a positive correlation with the stress concentration of nTiO2 (ranged 5~15 mg/mL); then total RNA of P. polycephalum was isolated from the stressed P. polycephalum. The further findings were the transcriptional responses to the stress of nTiO2 of glutathione s-transferase gene (Ppgst), and there existed a positive correlation between the transcriptional level and nTiO2 exposure concentrations within a range (5~15 mg/mL). In conclusion, Ppgst could be used as a kind of biomarker to indicate the environmental pollution of nTiO2 of different diameters.
