Isolation, Identification and Characterization of a Decolorization Strain for Azo Dye Reactive Black 5

Abstract:A bacterial strain,EWA2,with remarkable ability to decolorize azo dye Reactive Black 5,was isolated from activated sludge collected from a wastewater treatment plant in Xi'an.By means of biochemical test and homology comparison of 16S rDNA sequence,strain EWA2 was identified as Proteus mirabilis.In LB liquid medium,decolofization rate of Reactive Black 5 (≤300 mg/L) by strain EWA2 could roached 94.7%~99.9/% within 48 hours.In simulated wastewater containing Reactive Black 5,strain EWA2 could utilize sodium carboxymethyl cellulose and glucose as carbon source to significantly increase the decolorization rate.Under the condition in 8% NaC1,the decolorization rate was 83.6% within 48 hours.It indicated that biodegradation was the main way of decolorizing Reactive Black 5 by strain EWA2,and the adsorption peak of degradation products appeared in near-ultraviolet region.
