Study on Screening and Expression of Metabolism-related Genes with Fish Dietary Exposure to Chemicals

Abstract:Experiments were carried out on the dietary exposure of Gobiocypris rarusto three substances,i.e.estradiol (E2),muskxylene(MX) and hexachlorobenzene (HCB),in terms of gene response,expression trends and characterization in different time points by using Agilent Zebrafish Oligo Microarray (4×44 K).Four metabolism-related genes including gla,ND4,psph and gss were screened out and their expression trends were coincident with the metabolic status of metabolizable substances such as MX and E2.Real time fluorescent quantitative-PCR (qRT-PCR) was used to check the real expression of these four genes in Gobiocypris rarus.Furthermore,an investigation was performed to compare the expression trends and levels of those four genes in Gobiocypris rarus and Danio rerio with the dietary exposure to MX,and the results suggested that expression trends and characterization were similar in two species of fish;and the fold-changes of gene expression levels of three up-regulated genes,such as gla,ND4 and gss,were higher than 1.5 in the early stage of exposure (after 3 or 4 days),which might reveal fish's metabolic abilities of chemicals in the early stage so the test and estimation for bioaccumulation of chemicals in fish could be simplified and optimized.
