Extraction Methods and Corrosion Characterization of Extracellular Polymeric Substances from Strain Attached on the Reclaimed Wastewater Pipelines

Abstract:Extracellular polymeric substances (EPS) play an important role on biocorrosion.The suitable extraction method for EPS with advantages of low cell lysis and no chemical extractants required that might otherwise disturb electrochemical process is helpful to explore the biocorrosion process.Microorganisms were cultured in the R2A medium with inocula taken from biofilm in a reclaimed wastewater distribution system and then EPS were extracted from the culture by six methods.The optimal extraction method was determined and then physicochemical characterization of EPS was analyzed.The results showed that water bath (60 ℃,40 min) was the best method for extraction of EPS.EPS extracted from different culturing stages contained different proportions of protein and polysaccharide but with similar functional groups.The production of EPS achieved the highest value at the stationary stage.EPS formed a protective film and retarded the cathodic reduction of corrosion while EPS also promoted anodic dissolution of cast iron through functional groups.
