Abstract:Soil samples were collected from different soil layers of oil contaminated site to study alkane content, the variation of microbial communities and activity of microbial metabolism, and the results showed that the oil contaminated soil had high alkane content, which was negatively correlated with bacteria and fungi(r=-0.885,P<0.05),and extremely significant negative correlated with actinomycetes (r=-0.948,P<0.01).Different soil layers of clean soil and oil pollution of bacteria, fungi and actinomycetes decreased with increasing depth, different soil layer of clean soil affected by oil stress after the number of bacteria and actinomycetes significantly reduced (P<0.01),the number of 0~15 cm soil fungi increased significantly (P<0.01),15~30 cm,30~45 cm soil layer did not increase significantly (P>0.01).Clean soil microbial metabolic activity was significantly higher than that of petroleum contaminated soil (P<0.01),and soil microbial metabolic activity increased with depth decreased.Utilization of six carbon sources is different for different soil layers of clean soil and oil contaminated soil.Principal component analysis (PCA) showed that different layers of clean soil and oil pollution of soil microbial community had significant difference in 31 carbon sources in the PCl direction (P<0.01),different soil layers is mainly reflected in the PC2 direction.Main difference of PC1,PC2 is I-erythritol,L-threonine in carbon source.
