Advances in multiple mechanisms of thrombospondin-1 in regulating tumour angiogenesis

Abstract:Thrombospondin-1(TSP-1)is a multifunctional extracellular matrix protein that plays a complex dual role in tumor angiogenesis.Recent research has made significant progress in elucidating the mechanisms by which TSP-1 both inhibits and promotes angiogenesis.TSP-1 exerts its regulatory effects for tumor angiogenesis primarily via interactions with cell membrane receptors like CD36,CD47,and integrins.The CD36-mediated signaling pathway inhibits tumor angiogenesis and induces endothelial cell apoptosis,whereas TSP-1 binding to integrins can potentially stimulate neovascularization.Additionally,TSP-1 influences the tumor microenvironment via activating the transforming growth factor-beta(TGF-β)signaling pathway and modulating the activity of matrix metalloproteinases(MMPs),further affecting the dynamic balance of tumor angiogenesis.Due to its multifaceted functions,TSP-1 exhibits distinct biological effects in different tumor types and disease stages.A deeper understanding of its mechanisms is critical for deciphering the key regulatory networks of tumor angiogenesis and can provide novel therapeutic strategies targeting TSP-1-mediated pathways.Future studies should further explore the tissue-specific roles of TSP-1 and integrate molecular targeting approaches to enhance the precision and efficacy of cancer therapies.
