Advances of Src protein kinase in liver cancer

Abstract:Primary liver cancer(PLC)is one of the most prevalent and lethal malignancies worldwide,characterized by complex pathogenesis and poor prognosis.Src is the first identified transformation product of oncogenic viruses and the prototype of SRC family kinase(SFK),which plays a critical role in cell growth,division,migration and survival signaling pathways.It is involved in several signaling pathways such as mitogen-activated protein kinases(MAPK),phosphatidylinositol 3-kinase(PI3K)/protein kinase B(AKT1),signal transducers and activators of transcription(STATs),and closely related to the development of various tumors such as hepatocellular carcinoma.Aberrant activation of Src in PLC promotes metastasis,inhibits apoptosis,and enhances energy metabolism,thereby accelerating tumor progression and correlating significantly with unfavorable clinical outcomes.Furthermore,Src contributes to cancer-induced bone pain by modulating osteoclast activity and the tumor microenvironment,facilitating bone metastasis.This review systematically summarized the structural features and biological functions of Src,as well as its associated signaling pathways,with a focus on elucidating the molecular regulatory mechanisms of Src in the pathogenesis of PLC.The findings may provide novel theoretical insights for the development of targeted therapeutic strategies against PLC.
