Expression of lncRNA WDFY3-AS2 in gastric cancer and its effect on biological behavior of gastric cancer cells

Abstract:Objective To test the expression of long non-coding RNA(lncRNA)WDFY3-AS2 in gastric cancer tissues and their adjacent normal tissues,and to investigate its effect on the biological behavior of gastric cancer cells.Methods Forty gastric cancer patients hospitalized in the Gastrointestinal Surgery from January 2020 to December 2023 were enrolled.Gastric cancer and adjacent normal tissues were intraoperatively harvested to detect the expression of lncRNA WDFY3-AS2 by real-time reverse transcription fluorescent quantitative polymerase chain reaction(RT-qPCR).The gastric cancer cell line BGC-823 with low expression of lncRNA WDFY3-AS2 was selected for subsequent experiments.Cells were divided into the control group and the lncRNA WDFY3-AS2 overexpression group.Proliferation was detected by cell counting kit-8(CCK-8)assay and colony formation assay.Cell invasion and migration ability were detected by Transwell and cell scratch assays.Results LncRNA WDFY3-AS2 was significantly lowly expressed in gastric cancer tissue than adjacent normal tissue(P<0.05).LncRNA WDFY3-AS2 was significantly lowly expressed in gastric cancer cell lines BGC-823 and SGC-7901 than the normal gastric mucosal epithelial cell line(P<0.05).Overexpression of lncRNA WDFY3-AS2 significantly inhibited cell viability,colony formation ability,migratory cell number and wound healing rate than the control group(P<0.05).Conclusion LncRNA WDFY3-AS2 may be used as a promising therapeutic target for gastric cancer.
