Role and mechanism of NRF2 signaling pathway in Parkinson's disease

Abstract:Parkinson's disease(PD)is a chronic and progressive neurological disorder,mainly characterized by the loss of dopaminergic neurons and the abnormal alpha-Synuclein?(alpha-Syn)?aggregation.Recently,it has been shown that the nuclear factor erythroid 2 related factor 2(NRF2)signaling pathway plays an important role in the pathogenesis of PD.NRF2 is a transcription factor whose main function is to regulate the antioxidative stress response and cellular autophagy processes.In PD,activation of NRF2 signaling can reduce oxidative stress,suppresses inflammatory responses and neuronal damage,thereby protecting dopaminergic neurons from the toxic effects of alpha-Syn aggregation.Furthermore,several drugs and natural compounds have been found to treat PD by activating the NRF2 signaling pathway.In conclusion,the role of NRF2 signaling in PD is mainly reflected in terms of anti-oxidation,anti-inflammatory and neuroprotective,therefore,further studies of the regulatory mechanism of NRF2 signaling may provide new ideas and targets for the treatment of PD.
