Effect of etomidate on the malignant biological behavior of glioma cells by regulating the cAMP/PKA/CREB signaling pathway

Abstract:Objective To investigate the effect of etomidate(Eto)on the malignant biological behavior of glioma cells by regulating the cyclic adenosine monophosphate(cAMP)/protein kinase A(PKA)/cAMP-response element-binding protein(CREB)signaling pathway.Methods Glioma cells U87 were induced with blank control,low-dose,medium-dose and high-dose Eto(0.2,0.4 and 0.8μg/mL,respectively),and high-dose Eto+Sp-cAMPS(0.8μg/mL Eto plus 10μmol/mL cAMP activator).Cell proliferation was detected by cell counting kit-8(CCK-8)assay.Cell migration and invasion were examined by Transwell assay.Cell cycle and apoptosis were tested by flow cytometry.The enzyme-linked immunosorbent assay(ELISA)and Western blot were used to detect the expressions of proteins in the cAMP/PKA/CREB signaling pathway and apoptotic proteins.A tumor bearing mouse model was applied to verify the effect of Eto on U87 cell transplantation tumors.Results Compared with blank control,low-dose,medium-dose and high-dose Eto treatment significantly decreased the proliferative,migratory and invasive cell numbers,numbers of cells arrested in the S and G2/M phase,protein levels of cAMP,PKA,CREB and B-cell lymphoma protein-2(Bcl-2)and tumor volume and mass in mice,but increased numbers of cells in the GO/G1 phase,apoptotic rate and protein level of Bcl-2-associated X protein(Bax)in a dose-dependent manner(P<0.05).Compared with the high-dose Eto group,high-dose Eto plus Sp-cAMPS treatment significantly increased the proliferative,migratory and invasive cell numbers,numbers of cells arrested in the S and G2/M phase,protein levels of cAMP,PKA,CREB and Bcl-2 and tumor volume and mass in mice,but decreased numbers of cells in the G0/G1 phase,apoptotic rate and protein level of Bax(P<0.05).Conclusion Eto may inhibit the malignant biological behavior of glioma U87 cells by inhibiting the cAMP/PKA/CREB signaling pathway.
