Effect of etomidate on the apoptosis of neural cells induced by oxygen glucose deprivation/reperfusion by regulating the Wnt/β-catenin signaling pathway

Abstract:Objective To investigate the effect of etomidate(ET)on the apoptosis of neural cells induced by oxygen glucose deprivation/reperfusion(OGD/R)by regulating the Wnt/β-catenin signaling pathway.Methods OGD/R-induced N2B cells were treated with ET at varying concentrations of 0.5-10μmol/L,and the optimal concentration of ET was screened.N2B cells were treated with blank control,OGD/R,OGD/R+low-dose,medium-dose and high-dose ET,and OGD/R+high-dose ET+Wnt activator LiCL.Cell proliferation was detected by cell counting kit-8 assay.Apoptotic cells were observed by Hoechst staining,and apoptotic rate was quantified by flow cytometry.Relative levels of pro-inflammatory factors in cell supernatant were measured by enzyme-linked immunosorbent assay.Reactive oxygen species(ROS)was quantified by immunofluorescence.Western blot was performed to examine protein levels of Caspase-3,Wnt3a and β-catenin.Results ET treatment at the concentrations of 0.5-10μmol/L significantly promoted the proliferation of OGD/R-induced N2B cells.The concentrations of 1.25μmol/L,2.5μmol/L,and 5μmol/L were selected for subsequent experiments.Compared with those of blank control,OGD/R-induced N2B cells presented pyknotic nuclei,stronger staining,and significantly higher apoptotic rate,protein levels of Caspase-3,Wnt3a and β-catenin,ROS level,and supernatant levels of interleukin-1beta(IL-1β),IL-8,and IL-6(P<0.05).Compared with OGD/R-induced cells,those induced with OGD/R plus low-dose,medium-dose and high-dose ET had large and round cells,lighter staining,and significantly lower apoptotic rate,protein levels of Caspase-3,Wnt3a and β-catenin,ROS level,and supernatant levels of IL-1β,IL-8,and IL-6 in a dose-dependent manner(P<0.05).Compared to cells induced with OGD/R+high-dose ET,those induced with OGD/R+high-dose ET+LiCL had pyknotic nuclei,stronger staining,and significantly higher apoptotic rate,protein levels of Caspase-3,Wnt3a and β-catenin,ROS level,and supernatant levels of IL-1β,IL-8,and IL-6(P<0.05).Conclusion ET alleviates OGD/R-induced N2B cell apoptosis by inhibiting the Wnt/β-catenin signaling pathway.
