MiR-133a-3p affects the proliferation,invasion and metastasis of oral squamous cell carcinoma cells by regulating COL1A1

Abstract:Objective To investigate the potential role of microRNA(miR)-133a-3p in oral squamous cell carcinoma(OSCC)and the regulatory mechanism.Methods OSCC cells were treated with blank control,or transfected with miR-133a-3p overexpression plasmid,si-collagen type Ⅰ alpha 1 chain(COL1A1),COL1A1 overexpression plasmid,and miR-133a-3p overexpression plasmid plus COL1A1 overexpression plasmid.Real-time quantitative PCR(RT-qPCR)was used to detect the expression level of miR-133a-3p in OSCC tissues and cells.Cell proliferation,migration,invasion and apoptosis were examined by cell counting kit-8(CCK-8)assay,colony formation assay,Transwell assay and flow cytometry,respectively.An OSCC xenograft model in nude mice was created to explore the in vivo role of miR-133a-3p in the growth of OSCC.The hematoxylin and eosin(H&E)staining was performed to examine in vivo proliferation of OSCC.Dual-luciferase reporter assay and Western blot were performed to illustrate the regulatory effect of miR-133a-3p on COL1A1.Results Compared with paracancerous tissues and immortalized oral epithelial cell lines(HIOEC),miR-133a-3p in OSCC tissues and cells was significantly downregulated(P<0.05).Compared with the NC group,the cell viability,migration and invasion number of tumor cells in the miR-133a-3p group were decreased,and the level of apoptosis was increased(P<0.05).The cell viability,migration and invasion ability of the si-COL1A1 group were significantly decreased,while the apoptosis rate was significantly increased(P<0.05);Dual luciferase report confirmed that MiR-133a-3p targeted COL1A1 expression(P<0.05).In vivo data showed that miR-133a-3p significantly inhibited the growth of OSCC by targeting COL1A1(P<0.05).Conclusion MiR-133a-3p affects OSCC cell proliferation,migration,invasion and apoptosis by targeting COL1A1,which may be a potential prognostic biomarker in OSCC patients.
