Study on the expressions of CD55,CD59 and CD34 in patients with hematological diseases

Abstract：Objective To explore the diagnostic values of antigen expression rates and mean fluorescence intensities ofCD55,CD59 and CD34 in the early diagnosis and differential diagnosis of hematological diseases.Methods The expression levels and fluorescence intensities of CD55,CD59 and CD34 in peripheral blood,bone marrow and membrane protein were measured by flow cytometry in patients with hematological diseases and controls.Their correlations and specificities were identified.Results There were significant differences in the expression levels of CD55,CD59 and CD34 in peripheral blood and bone marrow between patients with paroxysmal nocturnal hemoglobinuria(PNH),myelodysplastic syndrome(MDS),aplastic anemia(AA),acute lymphoblastic leukemia(ALL),and healthy controls(P＜0.05).The expression levels of CD55,CD59 and CD34 were also significantly different among each type of hematological disease(P＜0.05).There were significant differences in the positive rate of CD55 and CD59 on membrane protein antigens among each type of hematological disease(P＜0.05),while that of CD34 was comparable(P＞0.05).The percentages and mean fluorescence intensities of CD55,CD59 and CD34 in red blood cells,monocytes and neutrophils were significantly different(P＜0.05),and the mean fluorescence intensities of CD55 and CD59 in red blood cells and lymphocytes were also significantly different(P＜0.05).The sugar water test,Rous test,acid hemolysis test and free hemoglobin detection in each disease group were significantly different(P＜0.05),while the haptoglobin detection was not significantly different(P＞0.05).Conclusion The abnormal expressions of CD55,CD59 and CD34 in peripheral blood,bone marrow and membrane protein antigen,and their mean fluorescence intensities and abnormal routine experiment can be used as effective diagnostic indexes in the early diagnosis and differential diagnosis of hematological diseases.
