Vitamin C and E protect DEHP-induced cytotoxic damage to the pancreatic islet MIN6β cells

Abstract:Objective To explore the protective effects of Vitamin C(Vit C)and E(Vit E)protect di(2-ethylhexyl)phthalate(DEHP)-induced cytotoxic damage to the pancreatic islet MIN6β cells and the underlying mechanism.Methods MIN6β cells were induced with 0.1%DMSO,DEHP at 0.25,0.5,1.0,2.0 mmol/L,DEHP at 0.25,0.5,1.0,2.0 mmol/L plus 50 μmol/L Vit C and 50 μmol/L Vit E,and 50 μmol/L Vit C and 50 μmol/L Vit E for 16,48 and 72 h.Cell viability was measured by MTT assay.Apoptotic rate and cell cycle progression were examined by flow cytometry.Results Compared with DMSO-induced cells,cells induced with DEHP+Vit C+Vit E had no significant change in cell viability at 48 h,lower viability at 16 and 72 h,higher apoptotic rate and higher proportion of cells in the G0/G1 phase.Compared with DEHP-induced cells,cells induced with DEHP(0.25,0.5 and 1.0 mmol/L)+Vit C+Vit E had significantly higher viability at 16,48 and 72 h(P<0.05),and those induced with 2.0 mmol/L DEHP+Vit C+Vit E had significantly higher viability at 16 h(P<0.05),but no change at 48 and 72 h(P>0.05).Cells induced with DEHP+Vit C+Vit E had significantly lower apoptotic rate,lower proportion of cells in the G0/G1 phase and higher proportion in the G2/M phase(all P<0.01).Conclusion Vitamin C and Vitamin E have protective effects on cell proliferation inhibition,cell apoptosis and cell cycle arrest induced by DEHP.
