Quercetin alleviates polycystic ovary syndrome with insulin resistance in rats by mediating advanced glycosylation end products

Abstract:Objective To investigate the role of quercetin(QUE)in ameliorating polycystic ovary syndrome with insulin resistance(PCOS-IR)in rats by regulating the advanced glycosylation end products(AGEs).Methods Female Sprague-Dawley(SD)rats in 6 weeks old were randomly divided into the control group,PCOS-IR group,low-dose QUE(QUE-L)group,medium-dose QUE(QUE-M)group,high-dose QUE(QUE-H)group and metformin hydrochloride(MH)group,with 10 rats in each group.Except for those in the control group,rats in the remaining groups were subjected to PCOS-IR modeling by feeding high-fat and high-sugar diet combined with gavage of letrozole sodium carboxymethyl cellulose suspension.After modeling,QUE at varied doses and MH were given by gavage according to the groups.The hematoxylin and eosin(H&E)staining was used to observe the histopathological changes of ovary in rats.Serum levels of testosterone(T),estradiol(E2),luteinizing hormone(LH),follicle stimulating hormone(FSH),fasting insulin(FINS)and AGEs were determined by enzyme-linked immunosorbent assay(ELISA).Fasting plasma glucose(FPG)was detected and the homeostatic model assessment for insulin resistance(HOMA-IR)was calculated.Immunohistochemical staining and Western blot were used to detect the positive expression of AGEs and the receptor for AGE(RAGE)in rat ovarian tissues.Results Compared with rats of PCOS-IR group,rats in the QUE-L group,QUE-M group,QUE-H group and MH group had significantly lower number of follicles in ovarian tissue of rats,more compact structure of ovarian granulosa cells,lower serum T,LH,LH/FSH,FPG,FINS,HOMA-IR,positive expression rates of AGEs and RAGE in ovarian tissues,protein levels of AGEs and RAGE,but higher E2 and FSH(P<0.05).Serum AGEs were significantly lower in rats of QUE-M,QUE-H and MH groups than the PCOS-IR group(P<0.05).Conclusion Quercetin can inhibit glycosylation and improve the insulin resistance in rats with PCOS-IR by inhibiting AGEs and RAGE.
