Preparations of nano-ZnO and Ag-doped ZnO nanostructures and their inhibitory effects on the proliferation of human hepatoma cell

Abstract:Objective To investigate the preparation methods of zinc oxide(ZnO)nanoparticles and Ag-doped ZnO nanoparticles,and the inhibitory effect of the nanoparticles on hepatoma HepG2 cells in vitro.Methods ZnO and 0.5%Ag-doped ZnO nanoparticles were synthesized and characterized by X-ray diffraction(XRD)and field emission scanning electron microscopy(FESEM).ZnO nanoparticles and Ag-doped ZnO nanoparticles were applied to hepatoma HepG2 cells.The proliferation of HepG2 cells was detected by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT)assay.Results FESEM showed that the morphology of the synthesized ZnO and Ag-doped ZnO nanoparticles was hemispherical nanoparticles with a small amount of aggregation.The average particle size of ZnO nanoparticles was about 15-20 nm,and that of Ag-doped ZnO nanoparticles was about 20-30 nm.XRD patterns further confirmed that the synthesized nanoparticles were pure ZnO,and Ag particles existed at the grain boundaries of ZnO grains,rather than entering the lattice of ZnO nanoparticles.MTT assay showed that ZnO and Ag-doped ZnO nanoparticles inhibited the proliferation of hepatoma cells in a concentration-dependent manner.Silver-doped ZnO nanoparticles were more effective than bare ZnO nanoparticles in inhibiting HepG2 cells.Conclusion ZnO nanoparticles and Ag-doped ZnO nanoparticles can inhibit the proliferation of HepG2 cells,and Ag-doped ZnO nanoparticles are more effective than bare ZnO nanoparticles in inhibiting the proliferation of HepG2 cells.
