STIM1 promotes the proliferation of salivary gland adenoid cystic carcinoma cells via upregulating the c-Myc/RPL35-mediated ribosomal pathway

Abstract:Objective To explore the mechanism of STIM1 overexpression in promoting the proliferation of salivary adenoid cystic carcinoma(SACC)cells.Methods STIM1-OE and STIM1-Vector were transfected into SACC-83 cells.SACC-LM cells were transfected with small interfering RNA si-STIM1,showing a silenced stromal interaction molecule 1(STIM1).The effects of STIM1-OE and si-STIM1 on cell proliferation were detected by MTT assay,colony formation assay and soft agar colony formation assay.Differentially expressed genes between the STIM1-OE and STIM1-Vector groups were obtained by transcriptome sequencing,and possible signaling pathways were analyzed by R language.The protein levels of c-Myc,RPSA,MRPL11,FAU and RPL35 were detected by Western blot.Results Overexpression of STIM1 increased cell viability,colony number,and colony number in soft agar(P<0.05).However,knockdown of STIM1 decreased cell viability,colony number,and colony number in soft agar(P<0.05).Gene set enrichment analysis(GSEA)and KEGG pathway enrichment analysis showed that MYC-mediated ribosome pathway was abnormally activated after overexpression of STIM1.Further verification showed upregulation of c-Myc and RPL35,while protein levels of RPSA,MRPL11 and FAU did not change significantly(P<0.05).Conclusion Overexpression of STIM1 promotes the proliferation of SACC cells by activating c-Myc/RPL35-mediated ribosomal signaling pathway.
