Therapeutic effect of pachymic acid on chronic pelvic inflammatory disease in rats by regulating the HMGB1/TLR4 signaling pathway

Abstract:Objective To investigate the treatment of chronic pelvic inflammatory disease(CPID)in rats with pachymic acid(PA)and its impact on the high mobility group protein B1(HMGB1)/Toll like receptor 4(TLR4)signaling pathway.Methods A CPID rat model was constructed.Rats were randomly assigned into Control group,CPID group,low-dose PA group,medium-dose PA group,high-dose PA group,and high-dose PA+r-HMGB1 group.The enzyme linked immunosorbent assay(ELISA)was applied to detect serum levels of inflammatory factors such as interleukin-1β(IL-1β),interleukin-6(IL-6),and tumor necrosis factor-α(TNF-α).The uterine swelling rate of rats in each group was detected.The hematoxylin and eosin(H&E)staining was applied to observe the pathological morphological changes in rat uterine tissues.Immunohistochemistry was applied to detect the positive expressions of intercellular adhesion molecule-1(ICAM-1)and transforming growth factor-β1(TGF-β1).Western blot method was applied to detect the protein expressions of HMGB1 and TLR4.Results Compared with those of the Control group,CPID rats had severe uterine damage,dilation of the uterine cavity,obvious endometrial congestion and edema,glandular atrophy,epithelial cell degeneration,necrosis,shedding and proliferation,infiltration of a large amount of inflammatory cells,red blood cell exudation,higher contents of IL-1β,IL-6,TNF-α,obvious uterine swelling,and higher protein expressions of ICAM-1,TGF-β1,HMGB1 and TLR4(P<0.05).Compared with those of CPID rats,those treated with low-dose,medium-dose and high-dose PA showed alleviated uterine damage,gradual reduction in uterine dilation,less endometrial congestion and edema,increased glandular volume,obviously reduced inflammatory cell infiltration,lower levels of IL-1β,IL-6 and TNF-α,less uterine swelling,and downregulated protein expressions of ICAM-1,TGF-β1,HMGB1 and TLR4 proteins in a dose-dependent manner(P<0.05).Compared with those induced with high-dose PA,rats treated with high-dose PA+r-HMGB1 had severer uterine structure damage,enlargement of uterine cavity,endometrial congestion and edema,gland atrophy,inflammatory cell infiltration and red blood cell exudation,higher levels of IL-1β,IL-6,TNF-α,obvious uterine swelling,and upregulated protein levels of ICAM-1,TGF-β1,HMGB1,and TLR4(P<0.05).Conclusion PA exerts a positive therapeutic effect on CPID rats by inhibiting inflammatory response and fibrous tissue proliferation via suppressing the HMGB1/TLR4 signaling pathway.
