Effects of type 2 diabetes on expressions of EGFR and RAGE and prognosis of colorectal cancer

Abstract:Objective To detect the expression levels of epidermal growth factor receptor(EGFR)and receptor for advanced glycation endproducts(RAGE)in type 2 diabetes mellitus(TCM)combined with colorectal cancer(CRC)and CRC solely by immunohistochemistry;to explore the influences of type 2 diabetes mellitus(T2DM)on EGFR and RAGE expressions in CRC tissue;and to identify the correlation between pathological characteristics and the prognosis of CRC.Methods A total of 59 CRC patients who underwent surgical treatment in Shijiazhuang People's Hospital from January 2019 to March 2022 were selected.They were divided into observation group(T2DM combined with CRC,n=29)and control group(CRC,n=30).The expression levels of EGFR and RAGE protein in CRC tissues of observation group and control group were detected by ElivisionTM plus two-step immunohistochemical method,and the influence of T2DM on their expressions was analyzed.The relationship between them and clinicopathological features was analyzed.Patients in the observation group and control group were followed up for 36 months,and the survival between the two groups was analyzed by spearman correlation analysis.Results EGFR and RAGE were positively expressed in both the observation group and the control group.EGFR was mainly located in the membrane of CRC cells,and RAGE was mainly located in the plasma of CRC cells.The positive expression rate of EGFR was 69%in the observation group and 33.3%in the control group.The positive expression rate of RAGE was 72.4%in the observation group and 70.0%in the control group.Compared with the control group,the positive expression rate of EGFR was significantly higher in the observation group(P<0.05),but there was no significant difference in the positive expression rate of RAGE between the observation group and the control group(P>0.05).The lymph node metastasis rate of observation group was significantly higher than that of control group(P<0.05).EGFR and RAGE expressions were not correlated with the sex,age,differentiation,tumor size,depth of invasion and lymph node metastasis in the observation group and control group(P<0.05).The positive expression of EGFR was negatively correlated with the median survival(P<0.05).Conclusion The positive expression rate of EGFR in CRC tissue of CRC patients combined with T2DM is higher than CRC patients.T2DM combined with CRC is prone to lymph node metastasis.Positive expressions of EGFR and RAGE are negatively correlated with the postoperative survival of CRC.
