Hepatitis B virus X gene promotes the proliferation of hepatic cells by downregulating Caspase-1

Abstract:Objective To investigate the effect of hepatitis B virus X gene(HBx)on the proliferation of HepG2 cells mediated by Caspase-1.Methods The pcDNA 3.1 vector loading Hbx(pcDNA3.1-HBx)was transfected into HepG2 cells using lipofectamine,with those transfected with negative vector as negative control(NC).Cells were collected 72h after transfection.The expression of HBx was detected by reverse transcription-polymerase chain reaction(RT-PCR).The mRNA and protein levels of Caspase-1 were detected by quantitative polymerase chain reaction(qPCR)and Western blot,respectively.The proliferation of HepG2 cells transfected with pcDNA3.1-HBx and NC was assessed by CCK-8 assay.Results Transfection of pcDNA3.1-HBx was success with the verification by RT-PCR.The mRNA and protein levels of Caspase-1 were significantly lower in HepG2 cells transfected with pcDNA3.1-HBx than those transfected with NC.The proliferative rate was significantly higher in HepG2 cells transfected with pcDNA3.1-HBx than those transfected with NC.Conclusion HBx promotes the proliferation of HepG2 cells by downregulating Caspase-1.
