Effect of microscopes with different parameters on re-examined complete blood count peripheral blood cell re examination

Abstract:Objective To study the effects of different optical technical parameters of microscope field of views on the results of manual leukocyte count and platelet count by blood smear method,and to verify the accuracy of the formula for estimating platelet count,thus standardizing the parameters and methods of complete blood count(CBC)microscopy.Methods The number of leukocytes and platelets were counted by an automatic blood cell analyzer,modified Neubauer hemocytometer plate method and blood smear method.Among them,two fields of view of 18.0 and 26.5 were adopted in the blood smear method.The white blood cell count and platelet count were converted through formulas(r=2,r=10,r=15,and formula for estimating platelet count),which were then subjected to correlation analysis and rank sum test to compare with those measured by an automatic blood cell analyzer and modified Neubauer hemocytometer plate method.Results There were significant differences in the white blood counts with the fields of view of 18.0 and 26.5 measured by an automatic blood cell analyzer,modified Neubauer hemocytometer plate method and blood smear method(P<0.05).At r=10 and r=15,there was a significant difference in the platelet count measured by blood smear method with two fields of view(P<0.05).With the field of view of 18.0,there was a significant difference in the high value of platelet count measured by the automatic blood cell analyzer and blood smear(P<0.05).With the field of view of 26.5,there was a significant difference in the platelet count at r=2,r=10 and r=15 among the measurements by an automatic blood cell analyzer,modified Neubauer hemocytometer plate method and blood smear method(P<0.05).There was a significant difference in the low value of platelet count at the field of view of 18.0 between the measurement of the formula for estimating platelet count and that measured by the automatic blood cell analyzer and modified Neubauer hemocytometer plate method(P<0.05).There was a significant difference in the normal value of platelet count at the field of view of 26.5 between the measurement of the formula for estimating platelet count and that measured by the automatic blood cell analyzer and modified Neubauer hemocytometer plate method(P<0.05).No significant differences were detected in the remaining indexes(P>0.05).Conclusion Microscopes with different fields of view have a certain degree of influence on the white blood cell count and platelet count measured by blood smear method.Formulas for estimating CBC greatly solves the problem of re-examinations,which is worth promoting and applying.
