Expression and significance of cyclophilin B in cancer tissues and serum of patients with gastric cancer
Abstract:Objective To investigate the expression of cyclophilin B(CypB) in cancer tissues and serum of patients with gastric cancer, and to explore its clinical significance.Methods A total of 130 patients were divided into observation group(n=80) and control group(n=50). Paraffin sections were made from cancer tissues and paracancerous gastric mucosa tissues(over 5 cm away from edges of cancers) of all subjects and stained by immunohistochemistry to detect the expression levels of CypB in tumor tissues and paracancerous gastric mucosa tissues. Moreover the serum levels of CypB,CEA,IL-17 at 1d before treatment and at 1 month after treatment were detected by ELISA. In addition, the clinical pathological data of patients were also collected and the clinical staging was carried out according to TNM staging (7th version,2010 by UICC). Results The positive rates of CypB in cancer tissues were significantly higher than those in paracancerous gastric mucosa tissues (P <0.05). There were 11 cases of stageⅠ,21 cases of stage Ⅱ,42 cases of stage Ⅲ,6 cases of stage Ⅳ, respectively. The positive rates of CypB were increased in order of stageⅠto stage Ⅲ (P <0.05),but there were no significant differences in positive rates of CypB between stage Ⅲ group and stage Ⅳ group (P>0.05). The serum levels of CypB of patients before operation in observation group were significantly higher than those in control group (P<0.05). The serum levels of CypB were increased in order of stage I to stageⅣ(P<0.05). At one month after operation,the serum levels of CypB in observation group were decreased significantly, as compared with those in control group (P <0.05). Before operation,the serum levels of CypB in observation group were significantly higher than those in control group (P<0.05). After operation,the serum levels of CypB in observation group were significantly decreased, as compared with those before operation (P<0.05),however,there were no significant differences in serum levels of CEA and IL-17 between two groups (P>0.05).In addition the cancer tissues were closely correlated to the serum levels of CypB before operation(P<0.05), moreover,before operation the serum levels of CypB were closely correlated to those of CEA,IL-17(P<0.05).Conclusion The overexpression of CypB in serum and tumor tissues of patients with gastric cancer is significant in evaluating patient's prognosis.
