Correlation between the serum levels of HMGB1 and oxidative stress as well as inflammatory reaction in patients with different types of fatty liver diseases
Abstract:Objective To observe the levels of serum high mobility group box 1 (HMGB1), indexes of oxidative stress and levels of inflammatory factors in patients with different types of fatty liver diseases (FLD), and to explore the relationship between these factors and fatty liver disease as well as their clinical significance.Methods A total of 640 patients with FLD were divided into alcohol fatty liver diseases group (AFLD group,n =270) and non-alcohol fatty liver diseases group (NAFLD group,n =370),at the same time,the 300 healthy subjects were enrolled as control group. ELISA assay was used to detect serum levels of HMGB1,MDA,SOD,GSH,interleukins-8(IL-8),TNF-α、IL-6.Results The serum levels of TC,TG,LDLC,ALT,AST,and GLU in NAFLD group were significantly higher than those in control group,while the levels of HDLC were significantly lower than those in control group( P <0.05).However there were no significant differences in these indexes between NAFLD group and AFLD group ( P >0.05). Moreover the BMI in NAFLD group was significantly higher than that in AFLD group and control group ( P <0.05).The serum levels of MDA in NAFLD group and AFLD group were significantly higher than those in control group,however, the levels of SOD and GSH were significantly lower than those in control group( P <0.05),and variation of these indexes was more significant in NAFLD group. The serum levels of HMGB1, IL6,TNFα,IL8 in NAFLD group and AFLD group were significantly higher than those in control group ( P <0.05), and variation of these indexes was more significant in NAFLD group. In addition the serum levels of HMGB1 in NAFLD group and AFLD group were positively correlated with those of IL-6,TNF-α,IL-8,MDA,respectively( r =0.3976,0.4217,0.4981, 0.3124, 0. 3275, 0. 3976), however, which were negatively correlated with those of SOD, GSH, respectively( r =-0.3326,-0.3652,-0.2988, -0.3106,P <0.05).Conclusion The serum levels of HMGB1 are increased in FLD, which may be involved in occurrence and progress of FLD by regulating indexes of stress and inflammatory factors.
