Expression of TESTIN in nasopharyngeal carcinoma and its effects on the proliferation,epithelial-mesenchymal transition,invasion and metastasis of nasopharyngeal carcinoma cell line in vitro
Abstract:Objective To observe the expression of TESTIN in nasopharyngeal carcinoma(NPC),and to explore the effects of TESTIN overexpression on the proliferation,epithelial-mesenchymal transition (EMT),invasion and metastasis of nasopharyngeal carcinoma cell line in vitro. Methods The expressions of TESTIN in NPC tissue specimens and nasopharyngeal carcinoma cell line were detected by immunohistochemical assay,Real time-PCR,Western-Blot,respectively. After transient transfection of TESTIN into NPC cell line 5-8F by means of lipofectamine TM 2000,the expression of TESTIN was detected by Real time-PCR and Western-Blot.The effects of TESTIN overexpression on the proliferation,EMT,invasion and metastasis of nasopharyngeal carcinoma cells were detected by MTT,cell scratching assay and cell invasion assay, respectively.Moreover the effects of TESTIN overexpression on the expression of genes related with invasion and metastasis were detected by Western-Blot.Results The expression levels of TESTIN in NPC tissues were significantly lower than those in normal nasopharyngeal tissues(P<0.05).The expression levels of TESTIN in NPC cell line-CNE1,CNE2,C666-1,5-8F and 6-10B were significantly decreased,as compared with those in human normal immortalized nasopharyngeal epithelial cell line NP69 (P<0.05).However,the expression levels of TESTIN in 5-8F cells transfected by pcDNA3.1/TESTIN were significantly increased (P<0.05).The overexpression of TESTIN obviously inhibited the proliferation, EMT, invasion and metastasis of 5-8F cells,at the same time, which up-regulated the expressions of E-cadherin, however, which down-regulated the expressions of N-cadherin,Vimentin,MMP-2 and MMP-9 (P <0.05). Conclusion The expression levels of TESTIN are significantly decreased in human NPC tissue and cell lines,moreover, TESTIN overexpression can inhibit the proliferation, EMT,invasion and metastasis of 5-8F cells.
