Abstract:Objective To observe the effects of LncRNA RSU1P2 on cervical cancer cells in vitro. Methods The overexpression of RSU1P2 was performed by transfection plasmid in Caski,HeLa and C33A cells, then the effects of RSU1P2 on cell vitality were detected by MTT test and colony formation assay. Moreover the effects of RSU1P2 on cell metastasis and invasion ability were measured by Transwell, and its effects on cell cycle and cell apoptosis were determined by flow cytometry. The changes of EMT related protein were detected by Western Blot. Besides the effects of RSU1P2 on cervical cancer tissues of nude mice were detected in vivo. Results After overexpression of RSU1P2 in Hela and C33A cells, the cell abilities of proliferation, metastasis, invasion and VM were significantly increased (P<0. 05),and the cell proportion at phase G1,phase S and the number of apoptotic cells were significantly decreased (P<0. 05). As compared with those in control group, the average weight and tumor volume in nude mice with RSU1P2-A were increased by approximately 110% (P < 0. 05). Conclusion The lncRNA RSU1P2 can promote the pathogenesis and development of cervical cancer by regulating the distribution of let-7A and its target genes.
