Study on E-selectin gene polymorphism in different degree of coronary artery lesion
Abstract:Objective To investigate the serum levels of E-selectin and the correlation between the second exon A561C and the fourth exon G98T gene polymorphism of E-selectin and different degree of coronary artery lesion.Methods A total of 178 patients with coronary atherosclerotic heart disease who were diagnosed and treated in our hospital from Januaty 2015 to March 2017 were enrolled as coronary heart disease group (CHD group),meanwhile,the 162 health examination subjects were enrolled as control group.The E-selectin levels were detected by double antibody sandwich method,and E-selectin gene polymorphism distribution status was analyzed by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP).Results The serum level of E-se1ectin in CHD group were significantly higher than those in control group (P<0.01).There were significant differences in serum soluble E-selectin levels among CHD subgroups (P<0.01).Among the three CHD groups,the serum levels of E-selectin in acute myocardial infarction (AMI) group were significantly higher than those in SA group (P<0.05),but there were no significant differences between UA group and AMI groups (P>0.05).In CHD group,there was no significant difference between G98T genotype,GT and GG genotype of E-selectin (P>0.05).The A561C genotype comparison showed that the fasting serum levels of E-selectin of AC genotype were significantly higher than those of AA genotype (P<0.05).There was significant difference in GT and GG genotype distribution in G98T genotype of E-selectin between AMI group and angina pectoris group (P<0.05),The risk of AMI in patients with GT genotype was 2.356 times as high as that of GG genotype (OR=2.356,95%CI=1.020~5.442).There was significant difference in the distribution of AC and AA genotype of A561C between AMI group and angina pectoris group (P<0.05).The risk of AMI in patients with AC genotype was 2.558 times as high as that of AA genotype (OR=2.558,95%CI=1.160~5.641).Conclusion The different degree of coronary artery injury may affect E-selectin gene polymorphism,the G98T and A561C gene polymorphisms are correlated to pathogenesis of coronary heart disease,which may also influence the serum levels of E-selectin.
