Expression and clinical significance of miRNA-34a and miRNA-29b in non-small cell lung cancer

Abstract:Objective To investigate the expression and clinical significance of miRNA-34a and miRNA-29b gene in non-small cell lung cancer (NSCLC),and to explore the correlation between the expressions and clinical features as well as chemotherapy efficacy in patients with NSCLC.Methods Eighty patients with NSCLC who were diagnosed definitely in our hospital were served as experimental group,at the same time, the other 100 patients with lung benign diseases were served as control group.The expression levels of miRNA-34a and miRNA-29b gene were detected by RT-PCR, moreover,the expressions of miRNA-34a and miRNA-29b gene as well as the therapeutic effects of chemotherapy were retrospectively analyzed.Results The expression levels of miRNA-34a in experimental group were significantly higher than those in control group (P<0.05),however, the expression levels of miRNA-29b in experimental group were significantly lower than those in control group (P<0.05).The expression levels of miRNA-34a in adenocarcinoma group and in squamous-cell carcinoma group were significantly higher than those in the other types of lung cancers (P<0.05).Moreover the expression levels of miRNA-34a in adenocarcinoma group were significantly higher than those in squamous-cell carcinoma group (P<0.05), however, the expression levels of miRNA-29b in adenocarcinoma group and in squamous-cell carcinoma group were significantly lower than those in the other types of lung cancers (P<0.05),furthermore,which in adenocarcinoma group were significantly lower than those in squamous-cell carcinoma group (P<0.05).The expressions of miRNA-34a and miRNA-29b were closely correlated with lung tissue grade,TNM stage and lymph node metastasis (P<0.05).The effective rates of chemotherapy in patients with high-expression of miRNA-34a were significantly lower than those of patients with low-expression of miRNA-34a (P<0.05), moreover, the effective rates of chemotherapy in patients with low-expression of miRNA-29b were significantly lower than those of patients with high-expression of miRNA-34a (P<0.05).Besides the expression of miRNA-34a in lung cancer tissues was negatively correlated with that of miRNA-29b (r=-0.79,P<0.01).Conclusion The high-expression of miRNA-34a exists in NSCLC,however,the low-expression of miRNA-29b exists in NSCLC,and their expression levels contribute to the clinical stage,histological grade and lymph node metastasis evaluation,moreover, the patients with low-expression of miRNA-34a and high-expression of miRNA-29b have higher effective rates of chemotherapy,which is regarded as one of the indexes of benefiting from chemotherapy.
