Expression and clinical significance of stromal cell-derived factor-1 and nerve growth factor in gliomas
Abstract:Objective To investigate the expression and clinical significance of stromal cell-derived factor-1 (SDF-1) and nerve growth factor (NGF) in gliomas.Methods Eighty-six pathological specimens of human brain glioma that were collected in our hospital from January 2012 to February 2016 were enrolled as observation group,moreover,which was redivided into 4 subgroups according to different pathological grade,including 15 cases in grade Ⅰ group,24 cases in grade Ⅱ group,26 cases in grade Ⅲ group,21 cases in grade Ⅳ group,moreover,the other 84 cases of brain tissue specimens of patients with craniocerebral trauma were seclected as control group.The expression levels of SDF-1 and NGF mRNA were detected by Real-time quantitative polymerase chain reaction (RT-PCR), at the same time,the expression levels of SDF-1 and NGF protein were detected by immunohistochemistry.Results The expression levels of SDF-1 and NGF mRNA gene and protein in observation group were significantly higher than those in control group (P<0.05).The expression levels of SDF-1 gene,NGF mRNA and protein in grade Ⅳ group were significantly higher than those in grade Ⅲ group,which in grade Ⅲ group were significantly higher than those in grade Ⅱ group, moreover, which in grade Ⅱ group were significantly higher than those in grade Ⅰ group, besides,the expression levels of SDF-1, NGF mRNA gene and protein in glioma were increased with the increase of pathological grade (P<0.05).Moreover the expression levels of SDF-1 mRNA in gliomas were positively correlated to positive rates of protein (r=0.95,P<0.05),and the expression levels of NGF mRNA were positively correlated to positive rates of protein (r=0.96,P<0.05).Moreover the high expression of SDF-1 NGF was independent factors to predict poor prognosis of glioma (P<0.01),and the high expression of SDF-1 NGF was closely correlated to whole poor prognosis of patients with glioma.Conclusion The high expression of SDF-1 and NGF in gliomas has a certain role in promoting progression of disease,both of them are closely correlated with pathological grade,thus,both factors play an important role in pathogenesis and development of glioma,it is expected to provide new ideas for clinical detection and treatment of glioma.
