Effects of stem cell mobilization on TNF-α, IL-1β and pulmonary alveolar surfactant in rats with acute lung injury

Abstract：Objective To investigate the effects of bone marrow stem cell mobilization on tumor necrosis factor-α(TNF-α),interleukin 1β (IL-1β) and pulmonary alveolar surfactant in rats with acute lung injury (ALI).Methods Sixty clean grade healthy male SD rats were randomly divided into observation group and control group,with 30 rats in each group.The animal models with ALI were established in the rats of both groups.After modeling the rats in observation group were given recombinant human granulocyte colony-stimulating factor (rhG-CSF) by subcutaneous injection,however,the rats in control group were given the same volume 0.9% sodium chloride solution,then the 30 rats in each group were sacrificed on 3d,7d,14d after intervention,respectively,and the radial alveolar counts (RAC) at different periods and the ratio of wet/dry weight were detected by HE staining, the levels of IL-1β and TNF-α in bronchoalveolar lavage fluid (BALF) were measured by ELISA,and the expression levels of BrdU and pulmonary surfactant protein A (SP-A) were detected by double immunofluorescence staining.Results The ratio of wet/dry weight and the levels of IL-1β and TNF-α on 3d,7d,14d after lung injury in observation group were significantly lower than those in control group (P<0.05),however, The RAC values on 3d,7d,14d after lung injury in observation group were significantly higher than those in control group (P<0.05).The BrdU and SP-A double positive cells labeling rates on 3d,7d,14d after lung injury in observation group were (0.74±0.13)%,(1.40±0.23)%,(2.33±0.31)%,respectively,which were significantly higher than those in control group (P<0.05).Conclusion The bone marrow stem cells mobilization has a protective effect on ALI,and its protective effect may be involved in multiple mechanism.
