Study of effects of Xiaoyao pill on ovarian oxidative damage and apoptosis of granulosa cells in androgen-induced sterile rats

Abstract:Objective To observe the effects of Xiaoyao pill (XYP) on ovarian oxidative damage and apoptosis of granulosa cells in androgen-induced sterile rats (ASR).Methods Forty female SD rats (9-day age) were randomly divided into control group (n=20) and model group (n=40).The rat models with ovulation failure (ASR) were established by subcutaneous injection with testosterone propionate,after modeling,the 40 rats were randomly divided into model control group and Xiaoyao pill treatment group (medication for 5 weeks),with 20 rats in each group.Then these rats were sacrificed and blood specimens were taken,the levels of T-AOC in serum and ovarian tissue were detected by colorimetry,and the levels of MDA in serum and ovarian tissue were detected by penthiobarbital method (TBA), and the expression levels of Caspase-3 protein were detected by immunohistochemisty.Results As compared with those in control group, the levels of T-AOC in serum and ovarian tissue in model group were significantly decreased,however,the levels of MDA were significantly increased (P<0.05).The expression levels of Caspase-3 protein in ovary granulosa cells were significantly higher than those in control group (P<0.01).As compared with those in model control group, the levels of T-AOC in serum and ovarian tissue in Xiaoyao pill treatment group were significantly increased,however,the levels of MDA were significantly decreased (P<0.05), moreover, the expression levels of Caspase-3 protein of ovary granulosa cells in Xiaoyao pill treatment group were significantly lower than those in model control group (P<0.05).Conclusion XYP can improve follicular development and ovulation failure.
