Experimental study on the protective effects of Naoxuetong tablets on cerebral ischemia in mice
Abstract:Objective To explore the protective effects and possible mechanism of Naoxuetong tablets on cerebral ischemia in mice.Methods The animal models were established including global brain ischemia models in mice, bilateral common carotid artery ligation and vagas nerve ligation models in mice,global brain ischemia-reperfusion (I/R) in mice, pulmonary embolism models in mice induced by collagen-epinephrine combination,and acute incomplete brain ischemia models in rats,then the protective effects of Naoxuetong low-dose group (NXT-L group) and Naoxuetong high-dose group (NXT-H group) on cerebral ischemia injury were observed.Results The lengthening survival time of decapitation gasping in mice of NXT-H group was (23.26±1.76)s,and the lengthening survival time after bilateral common carotid artery ligation and vagas nerve ligation in NXT-Lgroup and NXT-H group was (128.33±3.11)s,(1316±7.67)s,respectively.The levels of SOD were (150.02±18.51)U/mgprot in mice with global brain ischemia-reperfusion,which were increased by NXT-H,moreover, NXT-L and NXT-H decreased the MDA levels in mice with global brain ischemia-reperfusion by (26.80±1.92)nmol/mgprot,(26.43±1.90)nmol/mgprot,respectively.However NXT-L and NXT-H decreased the MDA levels in rats with incomplete brain ischemia-reperfusion by (6.36±1.67)nmol/mgprot,(5.78±0.90)nmol/mgprot,respectively.NXT-H could decrease cerebral index by (0.71±0.04)%,and the death number of mice+hemiplegia number (non-recovery) decreased by NXT-H were 11 mice (2+9) in mice models with pulmonary embolism induced by collagen-epinephrine combination.Conclusion NXT can produce protective effects on brain tissue of mice by increasing the activity of SOD, decreasing the production of MDA and reducing cerebral vascular permeability, moreover, which can decrease the death number of mice+hemiplegia number (non-recovery) in mice models with pulmonary embolism induced by collagen-epinephrine combination.
