Effects of ursodeoxycholic acid combined with Chinese medicines on bile acids, cholesterol anabolis m in rabbit models with cholecystolithiasis after minimally invasive cholecystolithotomy
Abstract:Objective To investigate the effects of ursodeoxycholic acid combined with Chinese medicines on bile acids, cholesterol anabolism in rabbit models with cholecystolithiasis after minimally invasive cholecystolithotomy .Methods Sixty male New Zealand rabbits were selected as research subjects , ang the animal models with cholecystolithiasis were established,then these rabbits underwent endoscopic minimally invasive cholecystolithotomy .The rabbits were divided into three groups according to different postoperative treatment modes :the rabbits in control group were given general diet ,the rabbits in model group were given lithogenic diet , and the rabbits in intervention group were given lithogenic diet +ursodeoxycholic acid combined with Chinese medicines .The serum levels of total cholesterol (TC), total bile acid (TBA), total bilirubin (TBIL), cholesterol 7-alpha hydroxylase (CYP7A1) and bile salt export pump (BSEP) transport factor acbc11 mRNA in gallbladder tissues were detected and compared among the three groups .Results After3 -week intervention and 5-week intervention, the serum levels of TC and TBIL in model group were obviously higher than those in control group , however,the serum levels of TBA were significantly lower than those in control group ( P <0.05).The serum levels of TC and TBIL in intervention group were significantly higher than those in model group , and the serum levels of TBA were significantlylower than those in model group ( P <0.05).The levels of CYP7A1 and acbc11 mRNA in gallbladder tissues of model group were significantly lower than those in control group ( P <0.05),which in intervention group were significantly higher than those in model group ( P <0.05).Conclusion The ursodeoxycholic acid combination with traditional Chinese medicines can improve the expressions of cholesterol 7-alpha hydroxylase and bile salt export pump transportation factor acbc 11 gene in gallbladder tissue, and regulate the metabolic levels of bile acid , cholesterol, bilirubin,therefor,which can prevent gallstones relapse after minimally invasive cholecystolithotomy.
