Effects ofβ-arrestin2 gene transfection on the proliferation of human pulmonary adenocarcinoma cells A549 in vitro
Abstract:Objective To observe the effects of exogenousβ-arrestin2 gene transfection on the proliferation of human pulmonary adenocarcinoma cells A549 in vitro.Methods The pcDNA3.1-β-arrestin2 plasmid and and empty carrier-pc DNA3.1 plasmid were introduced into A549 cells by liposomes transfection technique,then the cell clones that could express stably β-arrestin2 gene were established by G418 screening.The expression levels of β-arrestin2 were detected by RT-PCR and Western Blotting,respectively.The proliferation status of positive clones was observed by methythiazoletertraolium ( MTT) assay.Results The A549 cell line that could express stably β-arrestin2 gene was established by liposome transfection and screening.As compared with that in non-transfection group and blank carrier-transfection group,the growth velocity in the cells that were transfected byβ-arrestin2 gene was significantly decreased ( P <0.05).Conclusion The β-arrestin2 gene can inhibit the growth of of human pulmonary adenocarcinoma cells-A549 in vitro by inhibiting the activity of P65 protein.
