Effects of hTERT antisense oligodeoxynucleotide on the expression levels of Bcl-2 protein in endometrial carcinoma ;cells
Abstract:Objective To investigate the effects of hTERT antisense oligodeoxynucleotide ( ASODN) on the expression levels of Bcl-2 protein in human endometrial carcinoma Ishikawa cells.Methods Human endometrial carcinoma Ishikawa cells were cultured in the medium with 2.5, 5 and10 umol/L hTERT ASODN for 24h,48h,72h,then the cell proliferation of Ishikawa cells was detected by MTT assay.The apoptosis rate of Ishikawa cells and the expression levels of Bcl-2 protein were detected by flow cytometry on 48h.Results The hTERT ASODN inhibited proliferation of Ishikawa cells in a dose-dependent and time-dependent manner,moreover, which could induce apoptosis of Ishikawa cells in a dose-dependent way and could decrease the expression levels of Bcl-2 protein.Conclusion The hTERT ASODN can induce apoptosis of Ishikawa cells, inhibit tumor cell proliferation,and its action mechanism may be correlated to decreasing the expression levels of Bcl-2 protein.
