Experimental study on the therapeutic effects of autologous platelet rich plasma combined with low molecular weight heparin on inhalation pulmonary injury in rats
Abstract:Objective To investigate the therapeutic effects of autologous platelet rich plasma ( PRP) combined with low molecular weight heparin ( LMWH) on inhalation pulmonary injury in rats.Methods Forty-eight healthy rats ( SPF grade) were randomly divided into control group, model group, with 12 rats in each group.The animal models with smoke inhalation pulmonary injury were established in rats of model group, singleu-se group and combination group,and the rats in model group and control group were given 0.9% sodium chloride solution by intratracheal instillation , however,after smoke inhalation pulmonary injury the rats in single-use group were given PRP liquor by intratracheal instillation , but the rats in combinationgroup were given mixed liquor of PRP with LMWH by intratracheal instillation .Rseults The water content of lung tissues in model group, single-use group, combination group was significantly higher than that in control group ( P <0.05),moreover, which in combination group was significantly lower than that in model group and single-use group ( P <0.05).The serum levels of TNF-α,IL-6,IL-1β,PDGF in model group,single-use group, combination group were significantly higher than those in control group ( P <0.05),however, which in combination group were significantly lower than those in model group and single-use group ( P <0.05).The Brdu positive immunohistochemical labeling indexes in model group, singleu-se group, combination group were significantly higher than those in control group ( P <0.05), furthermore, which in combination group were significantly higher than those in model group and single -use group (P <0.05).Conclusion PRP combined with LMWH can obviously alleviate pulmonary edema and inflammation reaction in treatment of inhalation pulmonary injury in rats.
