The expression and clinical significance of low molecular weight protein tyrosine phosphatase in cervical carcinoma

Abstract:Objective To investigate the expression and prognostic value of low molecular weight tyrosine phosphatase(LMW-PTP)in cervical squamous cell carcinoma(CSCC). Methods One hundred and sixty cases of paraffin specimens of our hospital from March 2008 to August 2014 were enrolled in the study. These paraffin specimens were divided into control group,cervical intraepithelial neoplasia phase Ⅰgroup,phaseⅡ ~ Ⅲgroup and cervical squamous cell carcinoma group according to pathological changes degree. The levels of low molecular weight tyrosine phosphatase were detected by RT-PCR and immunohistochemistry. Results The expression levels of LMW-PTPmRNA in C33A,SiHa and CaSki cell lines were significantly higher than those in HaCaT cells( P ﹤ 0. 05),and the expression levels of LMW-PTP were the lowest in HaCat cells,as compared with those in the other groups( P ﹤ 0. 05). The expression levels of LMW-PTP in C33A,SiHa and CaSki cell lines were increased,however,the expression of LMW-PTP in HaCaT cells was weakly positive,which in C33A cells,SiHa cells,CaSKi cells was obviously increased. The expression levels of LMW-PTP were gradually increased during the changes from normal cervical squamous epithelium to CSCC,moreover,the expression levels of LMW-PTPmRNA in moderately differentiated CSCC tissues were significantly higher than those in well differentiated CSCC tissues( P ﹤ 0. 05),furthermore, which in poorly differentiated CSCC tissues were significantly higher than those in moderately differentiated CSCC tissues(P ﹤0. 05). Besides the expression levels of LMW-PTP in CSCC tissues were closely correlated to histological differentiation degree of patient with CSCC( P ﹤ 0. 05). Conclusion The low molecular weight tyrosine phosphatase plays an important role in the development of CSCC,which suggests that low molecular weight tyrosine phosphatase is helpful to improve prognosis of patients with CSCC.
