Cardioprotection effects of Ginsenoside Rg1 on ischemia/reperfusion injury in isolated hearts of rats
Abstract:Objective To investigate cardioprotection effects of Ginsenoside Rg1 on ischemia/ reperfusion( I/ R) injury in isolated hearts of rats,and to explore its action mechanism. Methods The Langendorff models of myocardial I/ R injury in rats were established. The 18 rats were randomly divided into three groups:control group,I/ R group and Ginsenoside Rg1 group,with 6 rats in each group. The rats’hearts in Ginsenoside Rg1 group were pretreated with Ginsenoside Rg1 100μmol/ L for 10min. The cardiac indexes including maximal rise/ fall of left ventricular pressure( ± dp/ dtmax)and rate pressure product(RPP)were measured,and the activities of creatine kinase(CK)and lactate dehydrogenase(LDH)were detected. The changes of phosphorylation of protein kinase B/ Akt,and its downstream target glycogen synthase kinase 3β(GSK-3β)and extracellular signal-regulated kinase1 / 2(ERK1 / 2)were detected after 10 - minute reperfusion. Results As compared with those in I/ R group,the activities of CK,LDH in Ginsenoside Rg1 group were significantly decreased and cardiac function was obviously improved( P ﹤ 0. 05 ),moreover,the expression levels of p-Akt and p-GSK-3βwere significantly increased( P ﹤ 0. 05). Conclusion Ginsenoside Rg1 can alleviate I/ R injury in isolated hearts of rats,and its action mechanism may be correlated to the activation of PI3K-Akt signaling pathway.
