Effects of grape seed procyanidin extract on the pathological morphous of atherosclerotic blood vessel in rabbits
Abstract:Objective To observe the effects of grape seed procyanidin extract(GSPE)on pathological morphous of atherosclerosis(AS)blood vessel in rabbits by means of established animal models with hyperlipidemia. Methods Forty-two New Zealand white rabbits were randomly divided into six groups:blank control group,hyperlipidemia model group,low-dose GSPE group(0. 1g/ kg),median-dose GSPE group(0. 2g/ kg),high-dose GSPE group(0. 4g/ kg)and lovastatin positive control group(1mg/ kg),with 7 rabbits in each group. The rabbits in blank control group were fed with normal diet,however, the rabbits in the other 5 groups were fed with high fat diet for 8 weeks to establish AS models,finally,the effects of GSPE on the pathological morphous of aorta endothelium injury of rabbits were observed and evaluated. Results As compared with that in hyperlipidemia model group,the aorta endothelium injury showed by general morphous,histology,scanning electron microscopy in median-dose GSPE group and high-dose GSPE group was improved at some extent,especially,the ratio of atherosclerotic plaque/ intima area was significantly decreased by 29. 5% ,27. 2% ,respectively( P < 0. 05). Moreover the ratio of intima/ tunica media was obviously decreased by 72. 5% ,60. 2% ,respectively( P < 0. 05). The good recovery and protective effects of GSPE on the ultrastructure of aortic tunica intima were observed in median-dose GSPE group and high-dose GSPE group,but there were no significant differences between low-dose GSPE group and hyperlipidemia model group( P >0. 05). Conclusion The high-dose and median-dose GSPE play a certain role in alleviating aortic tunica intima injury of rabbits with AS.
