Effects of hyperbaric oxygen post processing on inflammatory reaction caused by ischemia-reperfusion injury after skin flap transplantation in rats

Abstract:Objective To investigate the effects of hyperbaric oxygen( HBO)post processing on inflammatory reaction induced by ischemia-reperfusion( IR)injury after skin flap transplantation in rats. Methods Thirty-six male Sprague-Dawley(SD)rats were randomly divided into 3 groups:sham-surgery(SH)group,ischemia-reperfusion(IR)group and the HBO post processing ﹢ ischemia-reperfusion group,with 12 rats in each group. The animal models with abdominal pedicle skin flap were established,then the rats in HBO ﹢ IR group received HBO post processing. On the 8th day after surgery,the rats were sacrificed and the tissue specimens were collected at the proximal,intermediate and distal portions of the flaps. The tissue inflammatory infiltration was detected by HE staining and the expression levels of intercellular adhesion molecule-1( ICAM-1 ) and metalloproteinases-9( MMP-9 ) were detected by quantitative Real-time PCR( RT-PCR). Results HE staining showed that inflammatory cell infiltration in HBO ﹢ IR group was slighter than that at the same dissected position in IR group,moreover,the expression levels of ICAM-1 and MMP-9 in HBO ﹢ IR group were significantly lower than those in IR group( P ﹤ 0. 05). Conclusion Hyperbaric oxygen post processing may alleviate inflammatory reaction induced by IR injury after skin flap transplantation in rats through inhibiting the expression levels of MMP-9 and ICAM-1 so as to increase the survival rate of skin flap.
