Effects of oxymatrine combined with lamivudine on the expression levels of CD4+CD2+5 T regulatory cells, IL-10 and TGF-βin patients with chronic hepatitis B

Abstract:Objective To observe the changes of expression levels of CD4+CD2+5 T regulatory cells,IL-10 and TGF-βin patients with chronic hepatitis B ( CHB ) , and to investigate the effects of oxymatrine combined with lamivudine on the expression levels of these indexes.Methods One hundred and twenty patients with CHB who were diagnosed and treated in our hospital from January 2012 to January 2015 were served as treatment group,and the other 90 healthy subjects were served as control group.Treg proportion and CD4+T lymphocytes, the ratio of Treg/CD4+T lymphocytes in peripheral blood were detected by flow cytometry for both groups.The expression levels of FOXP3,IL-10,TGF-βwere measured by ELISA.The CHB viral load was detected by fluorescent quantitation PCR.The 120 patients with CHB were randomly divided into therapy group A ( group A) and therapy group B ( group B) ,with 60 patients in each group.The patients in group A,on the routine therapy, were given lamivudine, however, the patients in group B, on the basis of group A, were given oxymatrine, with a treatment course of 6 months for both groups.Then the changes of Treg,FOXP3,HBV-DNA,IL-10,TGF-βwere detected and compared between the two groups.Results As compared with those in control group, the Treg proportion, ratio of Treg/CD4+ T lymphocytes,expression levels of FOXP3,IL-10,TGF-βin CHB group were significantly increased ( P <0.05),moreover, Treg proportion,expression levels of FOXP3,IL-10,TGF-βwere positively correlated to ALT and viral load ( P <0.05).As compared with those before treatment, the Treg proportion, expression levels of FOXP3, HBV-DNA, IL-10, TGF-βwere significantly decreased after treatment ( P <0.05),furthermore, the decrease degree in group B was more obvious than that in group A ( P <0.05) .Conclusion The proportion of CD4+CD2+5 regulatory T cells, the expression levels of IL-10 and TGF-βare increased in patients with CHB, which may be involved in the pathological process of CHB through mediating immune inflammatory injury.Oxymatrine combined with lamivudine can play an effective anti-HBV role,and its action mechanism may be related to the decrease of CD4+CD2+5 regulatory T cells so as to improve the immune function of organism.
