The anti-inflammatory effect of fenofibrate and pioglitazone on adjuvant arthritis in rats

Abstract:Objective To investigate the anti-inflammatory effect of fenofibrate and pioglitazone on adjuvant arthritis (AA) in rats.Methods Twenty-four male Lewis rats were randomly divided into four groups .The rats in blank group and model group were treated orally with same doses of carboxymethylcellulose for 21 days,however, the rats in fenofibrate group or pioglitazone group were treated orally with fenofibrate 100mg-1 · kg-1 day-1 or pioglitazone 30mg-1 · kg-1 · day-1 for 21 days,respectively.The rat models with AA were established by caudal vein intradermal injection with complete Freund ’ s adjuvant 1mg/each rat.The severity of AA was evaluated by observing clinical indexes including body weight ,AA index score, swelling degree of heel .Bone mineral density ( BMD ) and femoral bone mineral content ( BMC ) were measured by dual-energy X-ray absorptiometry at day 20.The expression levels of IL-1β,IL-6 and IL-10 mRNA were detected by Real-time RT-PCR.Results As compared with those in model group ,the AA index score and swelling degree of heel were significantly decreased in fenofibrate group (100mg-1 · kg-1 · d-1 ) and pioglitazone group (30mg-1 · kg-1 · d-1 ) ( P <0.05), however, BMD and BMC were significantly increased ,so were the expression levels of IL-1β,IL-6 mRNA ( P <0.05),but the expression levels of IL-10 mRNA were obviously decreased ( P <0.05).Conclusion Both pioglitazone and fenofibrate have certain therapeutic effects on experimental adjuvant arthritis in rats.
