Influence of curcumin on the expression of MMP-2 mRNA and CD44v6 mRNA in human gastric cancer cells in nude mice model

Abstract:Objective To investigate the effects of curcumin on the expression of MMP-2 mRNA and CD44v6 mRNA in human gastric cells ,and to explore the possible action mechanisms of curcumin in inhibiting metastasis of gastric cancer . Methods The orthotopic transplantation tumor models were established in nude mice with SGC-7901 gastric cancer cell line , then the nude mice were randomly divided into control group and curcumin group , with 12 mice in each group .The tumor growth and metastasis condition were observed ,and the expression levels of MMP-2 mRNA and CD44 v6 mRNA in tumor tissue were detected by in situ hybridization .Results The successful rate of orthotopic transplantation tumor was 100%.The metastases of liver, peritoneal cavity and lymph node in curcumin group were significantly less than those in control group (P<0.05),moreover, the expression levels of MMP-2 mRNA and CD44v6 mRNA in curcumin group were significantly down-regulated,as compared with those in control group ( P <0.05).Conclusion Curcumin can inhibit gastric cancer metastasis in nude mice orthotopic transplantation tumor ,and its action mechanism might be related to down-regulation of the expression levels of MMP-2 mRNA and CD44v 6 mRNA.
