Study on the protective effects and action mechanism of oxymatrine on hypoxia and ischemia-induced arrhythmia in rabbit left ventricular outflow tract cells

Abstract:Objective To investigate the protective effects of oxymatrine on hypoxia and ischemia-induced electrophysiological abnormality of left ventricular outflow tract cells of rabbits, and to explore the action mechanism of oxymatrine in anti-arrhythmia.Methods The action potentials were recorded by conventional intracellular microelectrode technique and the amplitude of action potential (APA) and maximal rate of depolarization (Vmax),the 50%,80% of duration of action potential ( APD50 and APD80) and the rate of pacemaker firing (RPF) in rabbit left ventricular outflow tract pacemaker cells in normal control group, hypoxia and ischemia group, hypoxia and ischemia +oxymatrine group were observed.Results As compared with those in normal control group,the action potentials of APD50,APD80 of left ventricular outflow tract pacemaker cells in hypoxia and ischemia group were significantly decreased ( P <0.05),however,APA was obviously increased ( P <0.05);Vmax and RPF were significantly decreased( P <0.01), furthermore,there existed arrhythmia.The addition of oxymatrine (80μmol/L)in hypoxia and ischemia group could significantly prolong APD50,APD80 ( P <0.05); APA was decreased ( P <0.05);Vmax and RPF were significantly increased ( P <0.01).Twenty minutes after reperfusion,RPF recovered to normal levels.Conclusion The automaticity of left ventricular outflow tract pacemaker cells can be obviously influenced by hypoxia and ischemia,complicated by arrhythmia, however,oxymatrine can counteract the effects,and can decrease the electrophysiological abnormality induced by hypoxia and ischemia,which suggests that oxymatrine has obvious protective effects on hypoxia and ischemia-induced electrophysiological abnormality of left ventricular outflow tract cells of rabbits,and has obvious anti-arrhythmia effects.
