Effect of zonisamide on NO content in brain tissue of rats with epilepsy
Abstract:ObjectiveToinvestigatetheactionmechanismofzonisamideasanewantiepilepticdrug. Methods Twenty-four rats with epilepsy induced by pentrazol were randomly divided into three groups:zonisamide group, phenobarbital group, epilepsy model group, with 8 rats in each group,at same time, the other 8 normal rats were used as control group. The contents of NO,MDA and activities of NOS,SOD in brain tissues of the rats were detected for the four groups, meanwhile, the changes of hippocampus tissue structure of the rats were observed. Results The contents of NO,MDA and activity of NOS were significantly increased in epilepsy model group,as compared with those in control group ( P <0.05),however,the activity of SOD was significantly decreased in epilepsy model group ( P <0.05).The contents of NO,MDA and activity of NOS were significantly decreased in zonisamide group and phenobarbital group, as compared with those in epilepsy model group, however, the activity of SOD was significantly increased in both groups ( P <0.05).Furthermore,zonisamide could improve the structure of hippocampus tissue in rats with epilepsy. Conclusion Zonisamide plays an antiepileptic role by reducing the contents of NO in rats with epilepsy.
