Exp erimental study on the correlation betwe n chemokin e-CXCL12-CXCR4 and ovarian cancer in nude mi ec
Abstract:Objective To investigate the correlation between chemokine-CXCL12-CXCR4 and the pathogenesis of ovarian cancer.Methods The three kinds of ovarian cancer cells including SKOV3/CXCR4 transfected with plasmid, SKOV3/neg transfected with vector and SKOV3 which was not transfected were cultured in vitro.The 36 nude mice were randomly divided into three groups (12 mice in each group),and the peritoneal cavity transplanted tumor models in nude mice were established by intraperitoneal injection with the three kinds of ovarian cancer cells.Then the model mice in each group were subdivided into two groups on the second day after ovarian cancer cell inoculation: treatment group ( n =6 ) , control group ( n =6).The mice in control group were intraperitoneally injected with 0.9% sodium chloride solution,however,the mice in treatment group were intraperitoneally injected with CXCL12-CXCR4 antagonist-AMD3100.The transplanted tumor number and size in peritoneal cavity,ascites volume and mean survival time were observed after these mice died naturally. Results There were significant differences in body weight differences about 1 week after ovarian cancer cell inoculation between control groups and treatment groups ( P <0.05).There was a significant difference in mean survival time between SKOV3/CXCR4 control group and treatment group (CXCR4 antagonist-AMD3100) ( Z =-2.29, P <0.05),however, there was no significant difference in mean survival time between SKOV3/neg control group and treatment group ( P >0.05) , nor was between SKOV3 control group and treatment group ( P >0.05).There were significant differences in transplanted tumor mean number and weight in peritoneal cavity between SKOV3/CXCR4 control group and treatment group ( P <0.05) . Conclusion There is a correlation between chemokine-CXCL12-CXCR4 and pathogenesis of ovarian cancer,which may be one of main reasons in the pathogenesis of ovarian cancer,and may also reflect the degree of pathogenetic condition to some extent.
