Effect and action mechanism of AQP-5 on lymphatic metastasis of colorectal cancer
Abstract：Objective To investigate the expression of aquaporin-5 ( AQP-5) in colorectal cancer ,and to analyze the relationship between AQP-5 expression and lymphatic metastasis as well as prognosis of patients .At the same time,to detect the expression of MMP-2,MMP-9,TIMP-1,TIMP-2 in colorectal cancer in order to explore their significance .Methods The paraffin specimens of 98 cases of colorectal cancer tissues with positive lymph nodes metastasis and 40 cases of tumor adjacent tissues were collected .The expression levels of AQP-5 and MMP-2, MMP-9, TIMP-1, TIMP-2 proteins were detected by immunohistochemistry ( SP) ,moreover the patients ’ clinical data were recorded and the patients were followed up to analyze the correlation between AQP-5 and MMP-2, MMP-9, TIMP-1, TIMP-2 proteins Results The expression levels of AQP-5, MMP-2,MMP-9 in tumor tissues were significantly higher than those in tumor adjacent tissues ,however,the expression levels of TIMP-1,TIMP-2 in tumor tissues were significantly lower than those in tumor adjacent tissues ( P <0.05).The expression levels of AQP-5,MMP-2 were positively correlated to the numbers of lymph node metastasis ( P <0.05).Kaplan-Meier survival analysis showed that the survival time in patients with positive AQP-5 was relatively shorter ( P <0.05).The AQP-5 was closely related with MMP-2,MMP-9,TIMP-1in colorectal cancer tissues ( P <0.05).Conclusion AQP-5 can promote lymph node metastasis of colorectal cancer ,and positive AQP is a sign of poor prognosis .AQP-5 may regulate the expression of proliferation-related proteins to realize the effect .
