Expressions of multidrug resistanc-associat ed protein in peripheral blood mononuclear cells in patientswith breast cancer
Abstract:Objective To investigate the expression of multidrug resistance-associated protein ( MRP) in peripheral blood monouclear cells ( PBMCs) of healthy people and patients with breast cancer ,and to explore their possible roles in the pathogenesis of breast cancer .Methods The levels of TNF-α,IL-8 and IL-6 in serum were detected by ELISA in 50 patients with breast cancer ( patients group ) and 20 healthy subjects ( control group ) .After the PBMCs in both groups were extracted , the expression levels of breast cancer resistance protein ( BCRP ) and MRP2/3/4/5 mRNA were detected by Real-time quantitative RT-PCR and Western blotting .Results The levels of TNF-α,IL-8 and IL-6 in patients group were significantly higher than those in control group ( P <0.05 ).The expression levels of BCRP and MRP5 mRNA were significantly increased,as compared with those in control group ( P <0.05),however,there were no significant differences in the other kinds of MRP between two groups ( P >0.05).Conclusion The increase of proinflammatory factors in peripheral blood of patients with breast cancer may be correlated to the expressions of BCRP and MRP 5,however,the related mechanism needs to be studied further .
