Establishment of an animal model with cervical spondylotic myelopathy through bone cement compression nail
Abstract:Objective To establish an animal model of cervical spondylotic myelopathy (CSM) with typical clinical characteristics through bone cement compression nail in order to investigate the pathological changes of cervical spondylotic myelopathy and the changes in CT and MRI examination .Methods Twenty-five New Zealand white rabbits ( 12-week age ) were divided into two groups , experimental group ( n =20 ) and control group ( n =5 ) .Before operation all the animals underwent somatosensory evoked potential ( SEP) examination and motor function scoring .A bone cement compression nail was placed in front of C3 centrum of rabbits in experimental group .After operation the changes of nerve function were observed for 6 months,and SEP,CT imaging,MRI imaging were performed.Results The nerve symptoms appeared firstly during 3 months after operation and motor function showed obvious differences at 6 months after operation .The significant changes were found in amplitude and incubation period of SEP in experimental group ( P <0.05).The location of bone cement compression nail could be observed in sagittal view and axial view of CT in experimental group ,with average vertebral canal occupying rate being 40.2%.The spinal compression was observed in axial view image of MRI ,with average spinal compression rate being 43.9%in experimental group ,moreover ,the signal intensity in compression area in T 2 weighing sagittal view was obviously higher than that in non-compression area .Pathological examination showed that substantia grisea medullae spinaliskwtl anterior horn was flatten in experimental group , cornu anterius medullae spinalis cells were disappeared or necrosis , substantia alba medullae spinalis presented vacuole change , with axonal degeneration and slight demyelination changes , Furthermore substantia grisea medullae spinaliskwtl slight gliosis could be observed .Conclusion The animal model with cervical spondylotic myelopathy established by bone cement compression nail has typical clinical characteristics , which can be used in pathological examination,SEP,CT,MRI inspection,with simple making,good repeatability,which is a reliable model for researching related data of cervical spondylotic myelopathy .
