Effect of losartan on the expression of CGRP and ET-1in renal tissue of diabetic rats
Abstract:Objective To investigate the effects of losartan on the expression of CGRP and ET-1 in renal tissue of diabetic rats. Methods Forty-two rats were randomly divided into normal control group ,diabetes group and losartan treatment group.The diabetic models were established by intraperitoneal injection with STZ ,and the rats in losartan treatment group were given losartan 30mg/kg by gavage for 8 weeks.After 8 weeks,the urine and blood specimens were taken to be tested ,and the expression levels of CGRP and ET-1 protein in renal tissue of the rats were detected respectively by immunohistochemistry and semiquantitative RT-PCR.Results There were significant differences in 24h urine protein,serum creatinine (Scr),blood urea nitrogen (BUN) among the three groups ( P <0.05).The expression levels of CGRP protein and mRNA in renal tissue of diabetes group were significantly lower than those in normal control group ,however,the expression levels of ET-1 protein and mRNA in renal tissue of diabetes group were significantly higher than those in normal control group ( P <0.05).As compared with those in diabetes group the expression levels of CGRP in losartan treatment group were significantly increased ,however, the expression levels of ET-1 were significantly decreased ( P <0.05).Conclusion The losartan has protective effect on kidney through promoting synthesis of CGRP and reduce secretion of ET -1.
