Study on the mech anism of anti-i nflammatory effetc of imprvo ed Yuyangfan g on mouse modelw ith ulcerative colitis induced by dextran sulfate sodium
Abstract:Objective To investigate the mechanism of anti -inflammatory effect of improved Yuyangfang on mouse model with ulcerative colitis (UC) induced by dextran sulfate sodium (DSS).Methods Forty male C57BL/6 mice were randomly divided into four groups:normal control group , model group , treatment group , control group , with 10 mice in each group.Except for normal control group ,the mice in control group ,model group and treatment group were given orally 4%DSS to establish animal models with ulcerative colitis .After the models were successfully established ,the mice in control group and model group were treated with saline enema ,however ,the mice in treatment group were treated with Chinese medicine enema , in addition,the mice in control group were treated with salazosulfapyridine enema for 14 days.The expression level TNF-αin colon tissue were detected by immunohistochemisty .Results After treatment the treatment group was superior to control group and model group in the aspects of stools forming ,bloody stool,body weight lightening and general condition ( P <0.05).The expression level TNF-αin colon tissue of model mice with UC were significantly higher than those in normal control group ( P<0.05),however,which were obviously decreased in treatment group ,moreover,treatment group was superior to control group ( P <0.05).At the same time,the expression of TNF-αin mouse’s colon tissue of model group was mainly found in mucosa and submucosa tissue ,mainly concentrated on the tissues of erosion ,ulcer and small blood vessels dilatation ,TNF-αpositive cells in treatment group were significantly decreased ,as compared with those in model group and control group ( P <0.05). Conclusion The improved Yuyangfang is effective in treating ulcerative colitis in model mice ,and its action mechanism may be related to reducing TNF-αlevels and inhibiting inflammation reaction .
